N/B- Low 260/280 is a protein contamination while high 260/280 is RNA or residual organics contamination. Low 260/280 is organics and/or salt contamination.
The self-designed primer should be site-specify verified and accurate. It should be at least 50 nucleotides from target sequence if not 500 nt, with a length 18-30 nt, optimal being 20-25nt, G-C content 50-65% and Tm 55C-65C.
It shouldn’t hybridize on any level. Palindromes should be eliminated due to secondary structures formation.
